Microsatellite markers for the New Zealand endemic tree Fuchsia excorticata (Onagraceae)1

چکیده
UNLABELLED PREMISE OF THE STUDY Microsatellite markers were developed from a New Zealand endemic understory tree, Fuchsia excorticata, to investigate factors affecting the mating system. • METHODS AND RESULTS Using 454 pyrosequencing, 48 microsatellite markers were developed and tested for polymorphism within populations. Twelve of these microsatellite loci were identified as being polymorphic within at least three populations and consistently amplified in the four populations tested. These primers amplified di-, tri-, and tetranucleotide repeats with 1-10 alleles per population. • CONCLUSIONS These results indicate the utility of microsatellite loci for future mating system and population genetic studies in F. excorticata.

A comparison of the pigments in Fuchsia extorticata pollen with the pigments extracted from the unusually coloured blue-black “fuchsia” bee pollen confirms the New Zealand tree fuchsia as the source of the bee pollen. The pigments were identified as the p-trans-coumaroylated derivatives of delphindin-, petunidin and malvidin-3-O-glucosides and delphinidin-30-(/?-c/s-coumaroyl-glucoside). The an...
متن کاملProvenance Variation in Fuchsia (fuchsia Excorticata) in Relation to Palatability to Possums
Fuchsia (Fuchsia excorticata) has been heavily browsed and often killed by brushtail possums (Trichosurus vulpecula) in many New Zealand indigenous forests, but remains healthy at some sites despite long histories of possum occupation. To determine whether fuchsia varied genetically in its palatability to possums, material from six widely dispersed stands (provenances) was propagated, and leaf ...
متن کاملPhylogenetic relationships and biogeography of Fuchsia (Onagraceae) based on noncoding nuclear and chloroplast DNA data.
To examine relationships and test previous sectional delimitations within Fuchsia, this study used parsimony and maximum likelihood analyses with nuclear ITS and chloroplast trnL-F and rpl16 sequence data for 37 taxa representing all sections of Fuchsia and four outgroup taxa. Results support previous sectional delimitations, except for F. verrucosa, which is related to a Central American clade...

PREMISE OF THE STUDY Eleven nuclear and four plastid microsatellite markers were screened for two gypsum endemic species, Oenothera gayleana and O. hartwegii subsp. filifolia, and tested for cross-amplification in the remaining 11 taxa within Oenothera sect. Calylophus (Onagraceae). METHODS AND RESULTS Microsatellite markers were tested in two to three populations spanning the ranges of both ...
